Nothing seems blatantly wrong with your setup.

Have you ruled out network problems?  Can you telnet to .100 port 25 from 
.80?

Is .80's netmask anything funkier than 255.255.255.0?

Is .100 doing its own packet filtering?  Might it be ignoring packets from .80?

Not sure what the problem might be, but I don't sense that's it's a Zope issue.
